Understanding the Link Between Trauma and Substance Abuse

Lykos is essentially a corporate spinoff of the nation’s leading psychedelic advocacy group, the Multidisciplinary Association for Psychedelic Studies, or MAPS, which funded the studies. The group was founded in 1986 to promote the benefits of MDMA and other mind-altering substances. Women Veterans with PTSD PTSD and Alcohol Abuse were 67% more likely to initiate a guideline discordant medication. More than half of this effect was explained by bipolar disorder, age, and prior psychiatric medication. Military culture

However, the FDA meeting raised a number of ethical and safety issues about the two clinical trials that drug manufacturer Lykos Therapeutics used to make their case to the FDA’s Psychopharmacologic Drugs Advisory Committee. An independent group of experts expressed concerns that the data from clinical trials did not outweigh risks for treatment of post-traumatic stress disorder. Several comments about methodologic challenges in conducting these studies should be highlighted. The first issue is how to handle providing treatment of multiple psychiatric disorders in a safe and ethical manner.

A quarter of the sample reported that they were in a combat role in their parent unit. More than half of the sample reported at least one family relationship adversity in childhood and 17 % met the criteria for childhood antisocial behaviour. A fifth of the sample met the criteria for a CMD and 4 % met criteria for probable PTSD. Logistic regression was used to determine the association between alcohol misuse and probable PTSD, as well as between alcohol misuse and the three PTSD symptom clusters. Unadjusted and adjusted odds ratios (ORs) [adjusted for socio-demographic and military factors—sex, age, marital status, rank, last deployment and engagement type (regular/reservist)] were calculated.

Fifty-six per cent of the participants reported a positive history of driving under the influence of alcohol. Events that most frequently resulted in PTSD were torture (53%), being threatened with a weapon/kidnapped/held captive (39%), and sexual assault (37%). Whether the comorbidity between PTSD and AUD accompanies a neuroimmune profile that is predominantly proinflammatory in nature, and whether the added morbidity represents an aggravated proinflammatory state, remains unknown. Furthermore, it is unclear whether the correlates of comorbid PTSD in AUD are uniform across different countries and ethnicities. Tryptophan degradation along the kynurenine pathway by causing the release of neurotoxic metabolites is reported to be increased in stress-related psychiatric disorders [28]. Reduced neurogenesis and a lack of neurotrophic support, such as that reflected in reduced plasma brain-derived neurotrophic factor (BDNF) levels, as well as increased stress hormones are consistent findings in stress-related disorders, including PTSD [29, 30].
